How It Works

Polyglutamic acid is a chain of glutamic acid molecules — a polypeptide — produced by bacterial fermentation. It is the substance that makes natto stringy.

On the skin it works as a humectant. Humectants attract water molecules and hold them in the outer layer of the skin, which makes that layer swell slightly. Swollen surface skin is smoother, less flaky and reflects light more evenly, so fine dehydration lines look softer. Glycerin and hyaluronic acid do the same job by the same principle.

What is a bit different about polyglutamic acid is that it is a large molecule and it dries down into a thin, flexible film. That film gives it a mild secondary sealing effect — it slows water leaving as well as pulling it in — and it is why the finish feels slightly tacky and slightly plumping in a way that a thin hyaluronic acid serum does not.

On the marketing claim. You will see polyglutamic acid described as many times more hydrating than hyaluronic acid, usually with a specific multiplier attached. That figure is not something you can trace to convincing human data, and how much water a molecule holds in a test tube does not translate into how much it hydrates skin. The honest position is that polyglutamic acid is a legitimate humectant with a pleasant film-forming quality and comparatively little published human evidence behind it.

Dry skin is polyglutamic acid's clearest use. What distinguishes it from other humectants is the film it leaves behind, which gives it a mild secondary sealing effect and a slightly tacky, plumping finish. Published human evidence for it is comparatively slim, and how much water a molecule holds in a test tube does not tell you how it performs on skin.
Limited evidence
The film-forming behavior is why polyglutamic acid gets pulled into barrier repair marketing: it measurably slows water loss in the short term. That is a surface effect, not a rebuild, and there is little human evidence of it restoring barrier function. It works best layered under something occlusive rather than used alone.

How to Use It

Apply it to damp skin
  • This is the whole trick with any humectant, and the reason many people find them disappointing. A humectant applied to a dry face in dry air will pull some water out of the deeper skin rather than in from the environment, and skin can end up feeling tighter. Apply it to skin still damp from cleansing.
Always seal it
  • Follow with a moisturizer, or an emollient like squalane, or a heavier occlusive on very dry skin. The humectant brings water to the surface, the layer on top stops it evaporating. Neither step works well alone.
Where it goes in the routine
  • Early. After cleansing, before creams and oils. It is water-based and needs to reach the skin directly.

Pairing & Conflicts

What to pair it with

Hyaluronic acid and glycerin. Stacking humectants is genuinely sensible and most products do it. Glycerin is the cheapest and best-evidenced of the three, and a serum containing all three is a reasonable buy.

An occlusive or emollient on top — squalane, a ceramide cream, or petrolatum on very dry skin. This is not optional if you want the hydration to last.

Retinoids and acids. Polyglutamic acid does not interfere with them and helps offset the dryness they cause. Apply the active first on clean skin, then the humectant, then moisturizer — or use the humectant on alternate nights.

Niacinamide and ceramides in a barrier routine. No conflict.

What not to mix

There is no chemical conflict to report. Polyglutamic acid is inert and stable.

The mistakes are practical. Do not use it as your only moisturizer — a humectant with nothing over it does very little, and in dry air it can leave skin tighter than before. Do not apply it to bone-dry skin. Do not layer it under a heavy silicone product too quickly, or you will get pilling; let it dry for a minute first.

And do not buy it as well as a hyaluronic acid serum expecting the two to add up to something. They do overlapping jobs, and the money is better spent on what you put on top.

Side Effects

Polyglutamic acid is well tolerated. There is not much of a side effect profile, partly because it is mild and partly because it has not been studied heavily.

Common and expected
  • A slightly tacky or filmy feel, more noticeable than with hyaluronic acid.Pilling — small rolls of product — when layered under makeup or a silicone primer too soon. Skin feeling tighter rather than more hydrated, if it is applied without sealing it, or in very dry indoor air. Mild stinging on a broken or over-exfoliated barrier. Usually the barrier rather than the ingredient.
Tell your doctor
  • An itchy rash where you applied it. Allergy to polyglutamic acid itself appears to be rare, and other ingredients in the serum are likelier suspects.Skin that keeps feeling dehydrated despite using it daily. The routine, not the ingredient, is usually the problem — check that you are sealing it.
Stop and get care
  • Swelling, hives or difficulty breathing after use. This would be very unusual and would point to something else in the formula.

Who Should Avoid It

There is no group that needs to avoid polyglutamic acid on safety grounds. It is a mild, inert humectant with no known systemic effects, and it is fine in pregnancy and breastfeeding.

Two practical reasons to skip it.

If your skin is dry rather than dehydrated — flaky, rough and short of oil rather than short of water — a humectant serum is not what you need. An emollient or occlusive will do more. See dry skin.

If you already own a hyaluronic acid or glycerin serum you are happy with, adding this one is unlikely to be noticeable. They do the same job.

A note on soy. Polyglutamic acid is produced by fermenting bacteria, often grown on soybeans. The finished ingredient is a polypeptide of glutamic acid and is not soy protein, and topical reactions in soy-allergic people have not been a recognized problem. If you have a severe soy allergy, patch test.

If you have sensitive skin and have reacted to a serum containing it, the fragrance or preservative is a more likely culprit than the polyglutamic acid.

How Long Until It Works

Within minutes
Skin looks plumper and feels smoother. This is real, it is visible, and it is entirely surface hydration.
A few hours
That is roughly how long the effect lasts if you have sealed it with a moisturizer. Less if you have not, and less in dry air or air conditioning.
One to two weeks
With daily use, flaking and the tight feeling after cleansing settle. Makeup sits better. Fine lines that are really dehydration lines look softer.
Beyond that, nothing accumulates
Humectants do not build up an effect over months the way a retinoid does. What you get in week two is what you get in month six.
What it will not do
It does not treat wrinkles, acne, pigment or redness, and it does not repair a damaged barrier on its own — for that you need lipids as well, so see ceramides and damaged skin barrier.
When you stop
Skin returns to baseline within a day or two. There is no rebound dryness; the hydration simply stops being topped up.
